Job description

In this PhD project we aim a better understanding of the mechanisms involved in immunotherapy-related toxicity, with a focus on T-cell behavior. We will conduct a clinical study with translational research. We will use several innovative technologies in the domains of imaging, immunology, genomics, and pathology to obtain in-depth insights in the factors underlying the early development of immunotherapy-related toxicities. Will you join our team?

Immune checkpoint inhibitors, such as anti-programmed cell death-1 (PD-1), anti-programmed
cell death ligand-1 (PD-L1), and anti-cytotoxic T-lymphocyte antigen-4 (CTLA-4), have revolutionized the treatment of cancer in the past decades. Immune therapy resulted in overall survival benefit for patients with advanced stage cancer, and have become standard of care in several cancer types.

Immune checkpoint inhibitors release the brake of the immune system during priming of naive T-cells and during reactivation of memory anti-cancer T-cell responses. However, this systemic activation of immune cells and induction of potentially self-reactive T-cells also leads to off-target activity, so-called immunotherapy-related toxicity. Combined treatment with anti-CTLA-4 and anti-PD-1 antibodies leads to severe toxicity in more than half of the patients.

Tasks and responsibilities

  • Coordinate and control our site-specific immunotherapy-related toxicity trial (planning of study activities and to obtain biopsy material, blood, stool samples, etc).
  • Acquire knowledge of PET imaging and perform quantitative analyses.
  • Acquire knowledge of diverse laboratory techniques, including multi-dimensional flowcytometry and multi-colour immunohistochemistry, and perform quantitative analyses.
  • Actively participate in multi-disciplinary scientific meetings (laboratory meetings).

Department

The Department of Medical Imaging and department of Medical Oncology are part of the Radboudumc Center for Oncology. We emphasize on working in multidisciplinary teams to improve both patient care as well as research. The nursing department, day care and outpatient clinic are all integrated to guarantee a well streamlined process in the treatment for our patients.

Embedded in our academic patient care are early stage clinical trials, and translational research regarding pharmacology, pathology, genetics, radiology, nuclear medicine and tumour immunology. Both the department of Medical Imaging and department of Medical Oncology have a close relation with the department of Tumor Immunology focussing on translational research. The department of Tumor Immunology is embedded within the Radboud Institute for Molecular Life Sciences (RIMLS), a leading European research school that provides an outstanding, multi-cultural and thriving research setting within the Radboudumc.

The Radboudumc aims to excel in all academic tasks and in giving the patient an important voice in the treatment team. To act together within networks and partnerships are topical issues of the Radboudumc subjects for giving shape to an optimal organization of sustainable oncology care.

Research Institutes
At the moment there are more than 1,300 PhD candidates at our medical hospital. This number includes PhD candidates on our pay roll as well as external candidates (those employed somewhere else but researching on our premises).

  • Radboud Institute for Health Sciences: ± 700
  • Radboud Institute for Molecular Life Sciences: ± 400
  • Donders Center for Medical Neurosciences: ± 200
